Workshop Program

Scientific Research Center at the College of Science, is organizing a workshop on the principles of DNA sequencing technology on the 2nd and 3rd May 2018.

Objectives:

In this workshop, the participants will be trained according to an automated sequencing protocol on an ABI Genetic Analyzer 3130 with the following objectives:

  1. To provide the participants with all the necessary theoretical and practical background of
DNA sequencing to its full potential.
  2. Understanding how dideoxynucleotides can be used to obtain DNA sequence information
(Sanger  sequencing  method)  and  how  sequence  reading  can  be  automated  using
          fluorescent ddNTPs.
  3. Explaining why both strands of DNA are necessary to be sequenced and how sequencing
primers can be used to sequence a DNA fragment?
  4. Finally, to provide the participants with an overview, training in and discussion of basic
    sequence manipulation and analysis tools including;
    • Sequence assembly and editing.
    • Reading frames and coding region identification.
    • Pairwise and multiple sequence alignments.
    • Database searching, similarity and phylogenetic analyses. Workshop schedule:

This workshop will have 2 sections, the theoretical section and the practical section, over two successive days as follows:

Day 1:

9:00 - 9:15: Opening and welcoming speech by Prof. Dr. Jaladet M.S. Jubrael.

9:15 - 9:45: Introduction to the principles of DNA Sequencing and its applications. 9:45 - 10:00: Coffee Break

  1. The practical session for Day1 will include the following steps:

-     10:00 -10:45: Purification of PCR amplicon that will subsequently be sequenced.

-     10:45 -12:00: Gel Electrophoresis to assess the purified PCR product.

-     11:00-11:30: Reading frames and coding region identification

-     12:00-15:00: Preparation of BigDye sequencing reaction and Cycle sequencing.

Day 2:

-     09:00 - 12: Precipitation of Cycle sequenced reaction.

-     11:00-11:30: Pairwise and multiple sequence alignments.

-     12:00-14:00: Denaturation  & running samples in the ABI Genetic Analyzer 3130
